If you enable the SPAM BOX feature in your cpanel you need to maintain it.
SPAM BOX is only a holding facility until you can either check or delete the spam. If you don't periodically clear it out emails will accumulate and takeover your email account and hosting webspace.

Spam Box is currently: disabled
The SPAM BOX feature directs all emails marked as spam by spam assassin to that folder, periodically you need to check those emails and delete the ones you don't want, if you neglect this maintenance the SPAM BOX will continue to fill up and take your email account space/quota, once that quota is used any incoming emails will be rejected or terminated by the system until you empty the SPAM BOX.
You don't need SPAM BOX enabled for SPAM ASSASSIN to work properly.
SPAM ASSASSIN does its job by identifying SPAM and marking the offending email as SPAM, you can then setup a message rule in your email program on your computer to handle that MARKED message anyway you would like, you can have it sent to a certain folder for perusal, or straight to your TRASH, this is a better method as opposed to using SPAM BOX
If you are not sure how to setup a message rule in your email program on your computer you can always set Spam Assassin
to auto delete emails marked as spam
